Signalling and function of the human G protein-coupled receptor 55

چکیده
Results Here we show that GPR55 is activated by lysophosphatidylinositol (LPI), AM251, SR141716A (rimonabant) and AM281. GPR55 activation induces NFB, NFAT and CREB activation. Stimulation of GPR55 induces F-actin formation under the control of Ga13, RhoA and ROCK. We also show the suitability of Corning® Epic® DMR assay for GPR55 ligand screening. Furthermore, GPR55 activation leads to phosphorylation of extracellular signal-regulated kinase 1/2 (ERK 1/2).

G-protein Coupled Receptor Dimerization
A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...
متن کاملApelin: A promising therapeutic target? (Part 1)
Apelin is a recently discovered bioactive peptide, known to be an endogenous high-affinity ligandfor the previously orphan G protein-coupled receptor APJ. Apelin/APJ as a novel signaling pathwayhas been shown to play many crucial roles in cardiovascular function, blood pressure regulation, fluidhomeostasis, feeding behavior, obesity, type 2 diabetes mellitus, adipoinsular axis regulation, cellp...
متن کاملThe Effect of Aspartate-Lysine-Isoleucine and Aspartate-Arginine-Tyrosine Mutations on the Expression and Activity of Vasopressin V2 Receptor Gene
Background: Vasopressin type 2 receptor (V2R) plays an important role in the water reabsorption in the kidney collecting ducts. V2R is a G protein coupled receptor (GPCR) and the triplet of amino acids aspartate-arginine-histidine (DRH) in this receptor might significantly influence its activity similar to other GPCR. However, the role of this motif has not been fully confirmed. Therefore, the ...
متن کاملThe G protein-coupled receptor-associated protein 1 (GASP-1) regulates rimonabant-induced downregulation of GPR55
Background The G protein-coupled receptor 55 (GPR55) has recently been suggested to be responsible for those cannabinoid responses that could not be attributed to either the cannabinoid 1 (CB1) or cannabinoid 2 (CB2) receptor. Several potent GPR55 agonists were identified such as lysophosphatidylinositol (LPI) and several synthetic cannabinoids: One of these is rimonabant (SR141716A), an antago...
متن کاملG-protein-coupled receptors: evolving views on physiological signalling: symposium on G-protein-coupled receptors: evolving concepts and new techniques.
Introduction In 1967, Earl Sutherland described the simplest possible model for signalling through heptahelical receptors when he wrote, “It seems likely that in most, and perhaps all, tissues the receptor and adenylyl cyclase are the same” (Robison et al, 1967). Within ten years, Rodbell and Gilman had disproved this model by showing that heterotrimeric guanine nucleotide-binding proteins (G p...
